| I. Background | |
| IL-21 是 γc 家族細胞因子，主要由濾泡輔助性 T 細胞（Tfh）等 CD4 + T 亞群產(chǎn)生，通 過廣泛表達于 B 細胞、CD8+ T 細胞、NK 細胞的受體發(fā)揮作用。它在體液免疫中驅(qū)動生發(fā) 中心形成與高親和力抗體產(chǎn)生，在細胞免疫中維持 CD8+ T 細胞功能、防止耗竭，同時增 強 NK 細胞活性，是連接先天與適應性免疫的關鍵樞紐。其功能呈雙刃劍效應：過度激活 導致自身免疫病，信號不足則與慢性感染及腫瘤免疫逃逸相關。 IL-21 先與特異性 IL-21Rα 鏈結合，再募集共有 γ 鏈形成受體復合物，偶聯(lián)的 JAK1 與 JAK3 被激活，使受體胞內(nèi)段磷酸化并招募 STAT3。STAT3 磷酸化后形成同源二聚體入核， 調(diào)控 PRDM1、BCL6 等靶基因轉(zhuǎn)錄，介導 B 細胞漿細胞分化與 T 細胞干性維持。同時可輔 助激活 PI3K–AKT 等通路，信號強度受 SOCS 家族等負反饋精密調(diào)控。 |
