Three-prime repair exonuclease 1 (TREX1), a major endogenous 3′–5′ DNA exonuclease, is expressed in various cell types and functions as a homodimer. TREX1, encoding a 314-amino acid protein, is a single-exon gene located on chromosome 3p21.31. TREX1 has been intensively investigated for its ability to remove cytosolic DNA and avoid inappropriate innate immune activation .
