| KLK2 基因(激肽釋放酶 2)位于人類 19 號(hào)染色體短臂(19q13.4),編碼一種膜結(jié)合的胰蛋白酶樣絲氨酸蛋白酶,分子量約 51 kDa。其結(jié)構(gòu)包含兩個(gè)六股β桶和五個(gè)二硫鍵, 底物結(jié)合口袋 S1 帶負(fù)電,可與肝素等調(diào)節(jié)因子結(jié)合,增強(qiáng)與蛋白酶抑制劑的相互作用。 KLK2 基因的表達(dá)受雄激素受體(AR)嚴(yán)格調(diào)控,啟動(dòng)子區(qū)域含雄激素反應(yīng)元件(ARE), 在雄激素刺激下,AR 二聚化并轉(zhuǎn)位入核,結(jié)合 ARE 啟動(dòng)轉(zhuǎn)錄,因此 KLK2 特異性表達(dá)于前列腺上皮細(xì)胞及前列腺癌細(xì)胞,是前列腺癌的標(biāo)志物之一。 功能上,KLK2 能特異性切割 proPSA(前列腺特異性抗原前體)的 N 端肽鏈,激活其蛋白水解功能,參與精液液化及精子活動(dòng)調(diào)節(jié)。同時(shí),它通過激活 PSA 及促進(jìn)血管生成, 增強(qiáng)前列腺癌細(xì)胞的侵襲性和轉(zhuǎn)移能力,其高表達(dá)與腫瘤侵襲性呈正相關(guān)。 |
