| I. Background | |
| 趨化因子受體是 G 蛋白偶聯(lián)的七次跨膜細(xì)胞表面受體,其配體與其結(jié)合具有高親合力。迄今為止,至少有 20 種趨化因子受體已被確認(rèn),這些受體也可以分為四種亞型:CXC 趨化因子受體(CXCRs), CC 趨化因子受體(CCRs), XCR 和 CX3CR。 CXCR5(C-X-C 趨化因子受體 5 型)是特異性結(jié)合趨化因子 CXCL13 的 GPCR,核心功能為介導(dǎo) B 細(xì)胞、濾泡輔助 T 細(xì)胞(Tfh)及濾泡樹(shù)突細(xì)胞向次級(jí)淋巴器官(淋巴結(jié)/脾 臟濾泡)的定向遷移與定位,驅(qū)動(dòng)生發(fā)中心形成、抗體親和成熟及黏膜免疫應(yīng)答(如腸道派氏集結(jié)聚);其信號(hào)通過(guò) Gi 蛋白-PI3K/Rac 通路活化細(xì)胞骨架蛋白,建立免疫微環(huán)境空 間秩序。 |
